| 背景信息 | TNF receptor superfamily member 18 (TNFRSF18), also known as glucocorticoid-induced TNFR-related protein (GITR) or CD357, is a type I transmembrane protein that functions as a potent costimulatory receptor for T cells. It is constitutively expressed on CD25+CD4+ regulatory T cells and is rapidly upregulated on all T cell subsets following activation. Upon interaction with its ligand, GITRL, TNFRSF18 triggers signaling through TRAF-family members to activate the NF-kB and MAPK pathways. This signaling cascade promotes T cell activation, proliferation, and cytokine production while protecting cells from apoptosis. As an immune checkpoint molecule, TNFRSF18 is a significant target in oncology for enhancing antitumor immune responses. Additionally, its dysregulation is implicated in the pathogenesis of graft-versus-host disease and various autoimmune conditions, including rheumatoid arthritis, asthma, and atopic dermatitis. |
