| 背景信息 | Interleukin 1 receptor associated kinase 3 (IRAK3), also known as IRAK-M, is a pseudokinase that plays a critical role in modulating inflammation and immune responses. It is primarily expressed in monocytes and macrophages and is involved in the regulation of Toll-like receptor (TLR) signaling pathways. IRAK3 inhibits the dissociation of IRAK1 or IRAK2 from myddosome complexes, thereby downregulating NF-κB activation and reducing the production of inflammatory cytokines such as TNF-α and IL-6. IRAK3 is associated with endotoxin tolerance and sepsis, where its upregulation is linked to immunosuppression and worse clinical outcomes. Studies have shown that IRAK3 deficiency leads to enhanced sensitivity to TLR agonists and increased production of proinflammatory cytokines, highlighting its role as a negative regulator of inflammation. |
