| 背景信息 | Calpain 7 (CAPN7) is an atypical member of the calpain family of calcium-dependent, non-lysosomal cysteine proteases. Unlike classical calpains, it lacks the penta-EF-hand calcium-binding motif and instead features tandem microtubule-interacting and trafficking (MIT) domains. These domains enable CAPN7 to bind specifically to ESCRT-III proteins, such as IST1, facilitating its recruitment to midbodies and endosomes. CAPN7 plays a critical role in regulating membrane trafficking and signal transduction, specifically through the proteolytic activation required for cytokinetic abscission timing and the degradation of endosomal EGFR. It is ubiquitously expressed and is involved in diverse cellular processes, including the maintenance of the NoCut checkpoint and the modulation of cell migration. By integrating with the ESCRT machinery, CAPN7 ensures the precise execution of late-stage cell division. |
