| 背景信息 | Zinc fingers and homeoboxes 2 (ZHX2) is a nuclear transcriptional repressor belonging to the ZHX family, characterized by two N-terminal C2H2-type zinc finger domains and four C-terminal homeodomains. It functions by forming homodimers or heterodimers with ZHX1 and interacting with NF-YA to bind DNA and silence target genes. ZHX2 plays a vital role in regulating hepatic gene expression, specifically repressing fetal genes such as alpha-fetoprotein (AFP) and glypican-3 postnatally, while also influencing lipid and cholesterol homeostasis. Beyond its metabolic roles, it modulates NK cell maturation and retinal cell differentiation. In clinical contexts, ZHX2 exhibits dual roles in oncology; it acts as a tumor suppressor in hepatocellular carcinoma by suppressing AFP expression, yet functions as an oncogene in clear cell renal cell carcinoma and triple-negative breast cancer. |
