| 背景信息 | Lymphocyte antigen 96 (LY96), also known as Myeloid Differentiation factor 2 (MD-2), is a secreted protein that forms part of the lipopolysaccharide (LPS) receptor complex, which includes Toll-like receptor 4 (TLR4). LY96 binds bacterial LPS and facilitates its recognition by TLR4, triggering innate immune responses. Structurally, LY96 has a β-cup fold with a hydrophobic pocket for LPS binding, enabling dimerization of TLR4-LY96 complexes. This interaction activates signaling pathways such as NF-κB and MyD88-dependent Toll-like receptor signaling, leading to the production of pro-inflammatory cytokines and adhesion molecules essential for microbial defense. LY96 also cooperates with TLR2 to respond to components of Gram-positive and Gram-negative bacterial cell walls. Aberrant LY96 expression has been linked to inflammatory diseases like Crohn’s disease and rheumatoid arthritis. Additionally, LY96 overexpression in certain cancers, such as colon cancer, promotes tumor progression through inflammation-related pathways. |
