| 背景信息 | BUB1 mitotic checkpoint serine/threonine kinase (BUB1) is a central regulator of the spindle assembly checkpoint (SAC), essential for ensuring proper chromosome alignment and segregation during mitosis. It localizes to outer kinetochores where it recruits key checkpoint proteins, including Mad1, Mad2, and BubR1, and phosphorylates histone H2A to facilitate the recruitment of Shugoshin for centromeric cohesion protection. BUB1 also interacts with Cdc20 to regulate the timing of the metaphase-to-anaphase transition. While its catalytic activity supports SAC strength, the physical presence of the protein is critical for maintaining genomic stability. Clinically, dysregulation or mutation of BUB1 is well-linked to chromosomal instability and aneuploidy, which are hallmarks of cancer progression. Overexpression of BUB1 is frequently observed in various malignancies, including colon and lung cancers, where it serves as a driver of mitotic errors and a potential target for SAC-directed therapies. |
