| 背景信息 | PH domain and leucine rich repeat protein phosphatase 1 (PHLPP1) is a magnesium/manganese-dependent serine/threonine phosphatase that serves as a critical regulator of cell signaling. It specifically dephosphorylates the hydrophobic motif of various AGC kinases, including Akt2/3, PKC alpha/beta, and S6K1, as well as nuclear substrates like STAT1 and histones. By inactivating these targets, PHLPP1 functions as a broad tumor suppressor that dampens the PI3K-Akt and RAS-ERK pathways, thereby limiting cell proliferation and promoting apoptosis. It also modulates metabolic and immune signaling through the repression of receptor tyrosine kinase transcription. Clinically, the loss or dysregulation of PHLPP1 is linked to multiple cancers, including prostate and hematologic malignancies, and is associated with the development of obesity and type 2 diabetes due to impaired insulin signaling. |
