| 背景信息 | ATP binding cassette subfamily B member 1 (P-Glycoprotein) is a 170 kDa transmembrane glycoprotein that functions as an ATP-dependent efflux pump. It is a member of the ABC transporter family and plays a critical role in protecting tissues from toxic xenobiotics by expelling a broad range of substrates from the cytoplasm. P-Glycoprotein is highly expressed in physiological barriers, including the blood-brain barrier, intestines, liver, and kidneys. It operates through an alternating access mechanism, where ATP binding and hydrolysis drive conformational changes between inward-facing and outward-facing states to transport molecules across the membrane. Clinically, P-Glycoprotein is a major factor in multidrug resistance in various cancers, as it actively pumps out chemotherapeutic agents. It also significantly influences the pharmacokinetics and efficacy of drugs used to treat conditions such as epilepsy and HIV. |
