| 背景信息 | Protein tyrosine phosphatase non-receptor type 1 (PTP1B) is a ubiquitously expressed cytosolic phosphatase that localizes to the endoplasmic reticulum. It serves as a critical negative regulator of signaling pathways by dephosphorylating phosphotyrosine residues on receptor tyrosine kinases, most notably the insulin and leptin receptors. By attenuating these cascades, PTP1B acts as a metabolic brake, modulating energy homeostasis and cell growth. Functional studies indicate that the inhibition of PTP1B enhances insulin sensitivity and protects against diet-induced obesity. Clinically, PTP1B is a well-established candidate gene for type 2 diabetes and obesity, making it a significant target for therapeutic intervention in metabolic disorders. |
