背景信息Phosphatidylethanolamine binding protein 1 (RKIP) is a multifunctional protein that serves as a critical modulator of the MAPK/ERK and NF-kappaB signaling pathways. It functions through a phosphorylation-dependent molecular switch: in its unphosphorylated state, RKIP binds to Raf-1, preventing the activation of MEK and downstream ERK signaling. Upon phosphorylation by protein kinase C at Ser153, RKIP dissociates from Raf-1 and instead inhibits G protein-coupled receptor kinase 2 (GRK2), thereby enhancing GPCR signaling. RKIP also interacts with kinases such as TAK1 and NIK to regulate NF-kappaB activity. Clinically, RKIP is a well-established metastasis suppressor that is frequently downregulated in various cancers, including melanoma and prostate cancer. Its loss promotes tumor progression, invasion, and chemoresistance, and is also linked to inflammatory pathologies such as arthritis.
