| 背景信息 | AHNAK nucleoprotein (AHNAK) is a large structural scaffold protein that plays diverse roles in cellular organization, signal transduction, and membrane repair. It regulates cardiac calcium signaling by interacting with L-type Ca2+ channel beta subunits and the actin cytoskeleton to form subsarcolemmal networks. AHNAK also serves as a scaffold for key signaling enzymes, including phospholipase C-gamma and protein kinase C-alpha, and potentiates TGF-beta signaling through its interaction with Smad3. Beyond its structural functions, AHNAK is involved in blood-brain barrier formation and extracellular vesicle release. Clinically, AHNAK is associated with megacolon and neuroblastoma. It acts as a context-dependent regulator in oncology, functioning as a tumor suppressor through p53 modulation while also contributing to tumor metastasis and the maintenance of cancer stem cell properties. |
