| 背景信息 | Tripartite motif containing 40 (TRIM40) is an E3 ubiquitin-protein ligase that functions as a critical negative regulator of innate immune responses. It utilizes its tripartite motif domains to mediate the ubiquitination and proteasomal degradation of RIG-I and MDA5 receptors, thereby inhibiting RLR signaling. Additionally, TRIM40 promotes the neddylation of NEMO to stabilize NFKBIA, which effectively blocks NF-kappa-B activation and nuclear translocation. In the gastrointestinal epithelium, TRIM40 serves a protective role by suppressing inflammatory signaling and preventing carcinogenesis. It also interacts with ROCK1 to inhibit the proliferation of colorectal cancer cells via the c-Myc/p21 axis, positioning it as a key modulator of both immune homeostasis and tumor suppression in the gut. |
