| 背景信息 | WD repeat domain 59 (WDR59) is a signaling adaptor protein and an integral component of the GATOR2 complex. It features a characteristic WD40 domain that facilitates protein-protein interactions within the nutrient-sensing machinery of the cell. WDR59 plays a pivotal role in the amino acid-sensing pathway that regulates the activity of the mechanistic target of rapamycin complex 1 (mTORC1). When amino acids are abundant, WDR59 and the GATOR2 complex inhibit the GATOR1 complex, which otherwise acts as a negative regulator of mTORC1. This inhibition allows for the activation of mTORC1 at the lysosomal membrane, promoting cell growth and metabolism. Conversely, amino acid depletion leads to the suppression of this pathway. Clinically, WDR59 has been associated with epilepsy phenotypes, although the exact mechanistic link between its dysfunction and seizure pathogenesis is still under investigation. Its role in fundamental metabolic regulation underscores its importance in maintaining cellular homeostasis. |
