| 背景信息 | HECT, C2 and WW domain containing E3 ubiquitin protein ligase 1 (HECW1) is an E3 ubiquitin ligase that belongs to the NEDD4 family, containing C2, WW, and HECT domains. It is predominantly expressed in the central nervous system and plays a role in protein ubiquitination and degradation. HECW1 regulates various cellular processes, including protein trafficking, RNA metabolism, and autophagy/endolysosomal pathways. It can monoubiquitinate or polyubiquitinate substrate proteins, affecting their stability, localization, and function. HECW1 interacts with proteins involved in neuronal homeostasis and has been implicated in neurodegenerative diseases, particularly familial Amyotrophic Lateral Sclerosis (fALS). It has also been associated with colorectal carcinogenesis and non-small cell lung cancer metastasis. HECW1 expression is upregulated during neuronal differentiation and downregulated with aging, suggesting a protective role in neuronal homeostasis. |
