| 背景信息 | RUN and FYVE domain containing 3 (RUFY3) is a cytosolic adaptor protein that functions as an effector for small GTPases, including Rap2 and ARL8b. In neurons, RUFY3 localizes to growth cones and lipid rafts, where it is essential for establishing neuronal polarity and axon specification by organizing Rap2-dependent actin dynamics. It also acts on endolysosomes by recruiting the JIP4-dynein-dynactin complex to facilitate retrograde lysosome transport and perinuclear positioning. These roles allow RUFY3 to regulate both structural development and organelle distribution. Clinically, RUFY3 overexpression is linked to the progression of gastric cancer. It promotes tumor cell migration and invasion through PAK1-dependent pathways, suggesting a pro-metastatic role in various malignancies. |
