| 背景信息 | Leukocyte immunoglobulin like receptor B2 (LILRB2) is an inhibitory receptor predominantly expressed on myeloid cells, including monocytes, dendritic cells, and macrophages. It features four extracellular Ig-like domains and a cytoplasmic tail containing immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit phosphatases such as SHP-1 and SHP-2 to dampen immune signaling. LILRB2 binds a broad spectrum of classical and non-classical MHC class I molecules, including HLA-G, to promote immune tolerance. Beyond its role in the immune system, LILRB2 acts as a receptor for beta-amyloid oligomers, contributing to synaptic plasticity impairment in Alzheimer's disease. It is also implicated in the progression of acute myeloid leukemia, where it modulates the tumor microenvironment to favor immune evasion. |
