| 背景信息 | Family with sequence similarity 81 member A (FAM81A) is a brain-enriched postsynaptic scaffold protein localized to the postsynaptic density (PSD) of excitatory glutamatergic synapses. It plays a central role in organizing the PSD by facilitating the assembly of key components, including PSD-95, SynGAP, and NMDA receptors. FAM81A promotes liquid-liquid phase separation, which leads to the formation of membrane-less condensates that enlarge dendritic spines and support neuronal firing. The protein contains an SMC_N-like domain, suggesting potential interactions with nuclear or chromatin-associated proteins, possibly involved in synapse-to-nucleus signaling. While FAM81A is essential for normal neuronal activity and synaptic organization, its specific clinical associations remain under investigation. |
