| 背景信息 | Ring finger protein 217 (RNF217) is a member of the RING1-IBR-RING (RBR) ubiquitin protein ligase family that functions as an E3 ligase to regulate iron homeostasis. It mediates the proteasomal and lysosomal degradation of ferroportin (SLC40A1), the sole cellular iron exporter, particularly in macrophages and enterocytes. This process is modulated by hepcidin signaling and oxidative stress via the NRF2 pathway. RNF217 contains dual RING finger motifs and a transmembrane domain, allowing it to accept ubiquitin from E2 enzymes and transfer it to specific substrates. Additionally, it interacts with the anti-apoptotic protein HAX1, suggesting a role in apoptosis signaling. Dysregulation of RNF217 has been linked to pathological conditions including leukemogenesis and multiple sclerosis. |
