| 背景信息 | Discs large MAGUK scaffold protein 3 (DLG3) is a membrane-associated guanylate kinase (MAGUK) family protein that serves as a critical scaffold at excitatory synapses. It orchestrates the localization and stabilization of glutamate receptors, particularly NMDA receptors, through its multiple PDZ domains. DLG3 interacts with key synaptic proteins, including NETO1, GRIN2B, and SYNGAP1, while its guanylate kinase-like domain binds to DLGAP proteins to maintain postsynaptic architecture. These interactions are essential for synaptic plasticity and learning following NMDA receptor signaling. Additionally, DLG3 negatively regulates cell proliferation by interacting with the adenomatosis polyposis coli (APC) tumor suppressor protein. Clinically, mutations in the X-linked DLG3 gene are well-established causes of X-linked intellectual disability and intellectual developmental disorder 90, highlighting its fundamental importance in neurodevelopment and neuronal function. |
