| 背景信息 | Rho/Rac guanine nucleotide exchange factor 2 (GEF-H1) is a microtubule-associated factor that activates Rho family GTPases, primarily RhoA, by catalyzing GDP-GTP exchange. It is typically sequestered in an inactive state on microtubules and becomes activated upon microtubule depolymerization, thereby coupling microtubule dynamics to actin cytoskeleton remodeling and cell migration. GEF-H1 is involved in regulating cytokinesis, epithelial barrier function, and mechanotransduction. Pathologically, GEF-H1 is transcriptionally upregulated by oncogenic Ras/Raf/MEK/ERK signaling, supporting transformed growth in Ras-driven tumorigenesis. It also contributes to TNF-alpha-mediated barrier dysfunction and wound healing. In immune contexts, GEF-H1 modulates antigen presentation and tight-junction permeability, linking cytoskeletal regulation to inflammatory responses and barrier integrity. |
