| 背景信息 | SUMO peptidase family member, NEDD8 specific (SENP8) is a cysteine protease that plays a central role in the NEDD8 conjugation pathway. It is responsible for processing precursor NEDD8 into its mature form and deconjugating NEDD8 from targets such as cullins and Ubc12. By preventing aberrant hyper-neddylation, SENP8 ensures the proper activation of cullin-RING ligases, which are essential for proteostasis and G1/S cell cycle progression. In the nervous system, it regulates actin dynamics and Wnt/beta-catenin signaling to support neurite outgrowth and synapse maturation. Clinically, SENP8 deficiency is associated with impaired cell growth and neurodevelopmental deficits. It is also linked to the host response in rare viral encephalitides, such as Kunjin encephalitis and Kyasanur Forest Disease. |
